Company description
The research group of Prof. Steven Johnsen (see https://www.rbct.de/en/molecular-cancer-epigenetics) is part of the Robert Bosch Center for Tumor Diseases (RBCT). The RBCT is a newly founded non-profit translational cancer research institute at the Bosch Health Campus in Stuttgart, in direct proximity to the Robert Bosch Hospital (RBK). As a part of the National Center for Tumor Diseases (NCT) Southwest and the Comprehensive Cancer Center Tübingen-Stuttgart, the RBCT is dedicated to innovative cancer research and the development of new treatment strategies.

The Johnsen group has a particular and long-standing expertise in the elucidation of complex gene regulatory networks that control pancreatic cancer pathogenesis. In particular, we utilize state-of-the-art techniques such as various genome-/ transcriptome-wide sequencing approaches, high-throughput CRISPR/Cas9-mediated screening, multiplex immunofluorescence staining of tissues, single cell RNA-sequencing, as well as genetic engineering and other work on tumor cell lines, primary patient samples and translational tumor models. Our main research goal is to identify gene regulatory mechanisms that can be exploited as novel therapeutic targets to improve treatment response and pancreatic cancer patient survival.

Job description
1. Utilize modern sequencing-based molecular biology approaches (RNA-seq, ChIP-seq, HiChIP, Hi-C, PRO-seq, etc.) to test hypothesis and examine epigenetic and gene regulatory mechanisms controlling pancreatic tumor cell identity and therapy response.
2. Bioinformatic analysis and interpretation of data followed by hypothesis testing and generation.
3. Perform multiplex immunofluorescence staining to confirm gene regulatory mechanisms in primary tumor tissue using multispectral imaging with an Akoya PhenoImager.
4. Perform tissue-based preparation of histological samples (FFPE tumor material) for single cell RNA-sequencing using the 10X Genomics technology.
5. Perform advanced cell culture experiments to examine the effects of drug treatment in various translational model systems.
6. Participation in internal or external research collaborations.
7. Documentation of work procedures, data acquisition and analyses

Qualifications & Requirements
1. Applicants must have completed a Master of Science in a related field (biology, molecular biology, biotechnology or similar)
2. The applicant must have already secured or be able to secure external funding for their position (e.g., doctoral fellowship from DAAD, China Scholarship Council, or similar)
3. Experience with cell culture, transfections, etc.
4. Extensive molecular biology experience (real-time PCR, Western blot, etc.)
5. Strong interest in learning and gaining proficiency in bioinformatic analyses
6. Strong work ethic and high level of self-motivation
7. Independent, goal-oriented and structured way of working and ability to work in a team
8. Confident written and spoken English skills
Additional information
We offer you:
1. An exciting and international research environment
2. Excellent experience and expertise in modern epigenetic methods
3. Outstanding bioinformatic expertise
4. Extensive modern infrastructure for all methods related to the project
5. Working in a committed team, characterized by mutual appreciation and support
6. Proven track record of successfully supervising more than 20 doctoral students to completion
7. Long track record in gene regulation and cancer research (see https://www.ncbi.nlm.nih.gov/myn ... ibliography/public/)

This position is contingent upon the applicant having attained or attaining an externally funded fellowship for the duration of their project.
